Özet
Pharmacokinetics and pharmacodynamics are the basis of pharmacology. Pharmacodynamics is defined as how the drugs act to biological systems whereas pharmacokinetics is defined as the drug absorption, distribution, metabolism, and excretion (ADME). The pharmacokinetic factors to achieve a safer and effective therapeutic management of drugs in an individual patient is one of the most important issues of precision medicine. Primary goals of pharmacokinetics is to enhance the efficacy of the drugs and reduce the toxicity according to high plasma levels. There is a strong correlation between plasma drug concentrations and the efficacy and/or toxicity of the drugs and this concentrations can be altered according to pharmacokinetic parameters. A drug's effect is primarily related to its concentration at the site of action and can be regulated in a personalized based by following up the plasma concentrations of the drugs by therapeutic drug monitoring (TDM). Pharmacodynamic features of the drugs are also important to specify and target the role player molecules of the diseases. These role player molecules are the targets of the drug discovery strategies of the novel drug candidates. Over the last decade, especially the therapeutics related to immunotherapy and gene therapy are mainly focusing on the specific genes and proteins that play crucial roles in the disease progress.
