Objective: The hippocampal metaplasticity response indicates the altered ability of neurons to undergo synaptic changes based on their activity history. The difficulty in induction of long-term potentiation (LTP) and facilitation of long-term depression (LTD) in the hippocampus in old age has led researchers to the calcium hypothesis and it has been targeted to increase intracellular calcium levels as a result of increased gene expression of voltage-dependent calcium channels (VDCC) in hippocampal neurons in old age. The aim of this study was to investigate the impaired synaptic plasticity due to aging by administering a substance (Gabapentin, GP) that acts on the α2δ subunit of VBKK and to reveal the impairment of learning and memory performance by metaplasticity assessment.
Material and Method: In this study, 24 Wistar Albino male aged (15 months old) rats were used. The rats were divided into two groups as Aged+GP (AG, n=12) and Aged Control ( AC, n=12) groups. GP was added to the drinking water at 50 mg/kg/day for 8 weeks. Under anesthesia, rats were subjected to a protocol to induce metaplasticity response to the perforant pathway. Basal recording was taken and priming stimulus was low frequency stimulus (LFS). Afterwards, tetanization was induced with high frequency stimulation (HFS). Field potentials including excitatory postsynaptic potential (fEPSP) and population spike (PS) were recorded from the hippocampal dentate gyrus.
Results: EPSP values recorded from the dentate gyrus in GP-treated old rats were lower after DFU, YFU and during the maintenance period compared to non-GP-treated old rats (P<=0.03), whereas PS values did not differ statistically in the same periods (>=0.05). Conclusion: Metaplasticity is critical for normal function, preventing synapses from entering saturated LTP or LTD states, keeping them in a dynamic functional range. Elucidation of the role of voltage dependent calcium channels in metaplasticity may help to explain the causes of age-related cognitive changes.
