This study aimed to isolate vancomycin-resistant Enterococcus faecalis and Enterococcus faecium from fecal samples collected from healthy dogs, characterize the recovered isolates by phenotypic and molecular methods, and assess their genotypic relatedness using ERIC-PCR (Enterobacterial Repetitive Intergenic Consensus-Polymerase Chain Reaction). For this purpose, fecal samples were collected from 100 healthy dogs brought to veterinary clinics in Kayseri, Türkiye. Following enrichment in Enterococci Broth supplemented with vancomycin, the samples were inoculated onto Enterococcus Selective Agar and Bile Esculin Azide Agar and incubated aerobically at 37°C for 48-72 hours. At the end of the incubation period, suspected red and brown-black colonies were evaluated, and pure cultures were obtained on 7% sheep blood agar. For identification of the recovered isolates, phenotypic tests (Gram staining, motility, catalase, oxidase, lactose, and H₂S tests) and molecular analyses (genus- and species-specific PCR) were performed. The clonal relationships among the E. faecalis and E. faecium isolates were analyzed using ERIC-PCR. As a result of the present study, 41% of the 100 fecal samples were found to be positive for Enterococcus spp., and a total of 47 Enterococcus spp. isolates were recovered. In the phenotypic and molecular identification, six of the isolates were identified as E. faecalis and three as E. faecium. ERIC-PCR analysis revealed that three E. faecium isolates exhibited a single genotypic profile, showing clonal proximity. In contrast, E. faecalis isolates could not be genotyped by ERIC-PCR. In conclusion, it was determined that clinically significant vancomycin-resistant E. faecalis and E. faecium are present at a low rate in the gut microbiota of healthy dogs, and it was considered that monitoring the epidemiology and clonal dynamics of these two species at the molecular level in pet populations would be beneficial.
