Impact of Bacteria–Algae Co-Cultivation on Chlorophyll Pigment Production and EPS Formation

Özet

Bu tez çalı¸smasında, Chlorella vulgaris mikroalgi ile Bacillus subtilis ve Bacillus cereus bakterilerinin olu¸sturdu˘gu birlikteli˘gin, mikroalg fizyolojisi ve üretilen ekzopolisakkaritlerin (EPS) biyoaktif özellikleri üzerindeki etkileri detaylı bir ¸sekilde incelenmistir. Arastırma kapsamında, alg-bakteri birlikteliginin klorofil pigment sentezi üzerindeki uyarımı, üretilen EPS’lerin yapısal karakterizasyonu, antioksidan ve antimikrobiyal kapasiteleri ile A549 (akciger) ve DLD-1 (kolon) kanser hücre hatları ve sa˘glıklı insan fibroblast hücre hattı üzerindeki sitotoksik etkileri arastırılmıstır. Elde edilen bulgular, alg-bakteri birlikteli˘ginin fotosentetik pigment sentezini saf kültüre oranla yakla¸sık 6 kat artırdı˘gını ortaya koymustur. EPS miktar tayini sonuçlarına göre, en yüksek verim saf B. subtilis (501,4 mg/L) kültüründen elde edilirken, en dü¸sük verim saf C. vulgaris (35,8 mg/L) örne˘ginde saptanmı¸stır. Kimyasal analizler (HPLC), tüm EPS örneklerinde fruktozun en baskın monosakkarit oldu˘gunu, kızılötesi analizler ise protein varlı˘gını kanıtlayan amid I ve karakteristik glikozidik ba˘g titre¸simlerini do˘grulamı¸stır. Biyolojik aktivite testlerinde, antioksidan kapasite açısından C. vulgaris- B. subtilis ko-kültüründen elde edilen EPS en yüksek radikal giderme aktivitesini (%22,03) sergilemi¸stir. Sitotoksisite analizleri, alg ve bakteri EPS’lerinin birle¸siminin, saf formlara göre kanser hücrelerinin ço˘galmasını daha etkin belirgin bir ¸sekilde baskıladı˘gını kanıtlamı¸stır. Sonuç olarak, alg-bakteri etkile¸siminin sadece biyokütle verimini de˘gil, aynı zamanda üretilen biyopolimerlerin biyolojik olarak aktif kalitesini de "Biyoaktif ve terapötik potansiyel" yönde modüle etti˘gi belirlenmi¸stir

This thesis thoroughly investigates the effects of the consortium formed by the microalga Chlorella vulgaris and the bacteria Bacillus subtilis and Bacillus cereus on microalgal physiology and the bioactive properties of the resulting exopolysaccharides (EPS). Specifically, the research examines the stimulatory impact of the algal-bacterial co-culture on chlorophyll pigment synthesis, alongside the structural characterization of the extracted EPS. Furthermore, the study evaluates their antioxidant and antimicrobial capacities, as well as their cytotoxic effects on A549 (lung) and DLD-1 (colon) cancer cell lines. The findings revealed that the algal-bacterial consortium enhanced photosynthetic pigment synthesis approximately six-fold compared to pure cultures. Quantitative analysis of EPS production indicated the highest yield in the pure B. subtilis culture (501.4 mg/L), whereas the pure C. vulgaris culture exhibited the lowest yield (35.8 mg/L). Chemical composition analyses via HPLC confirmed fructose as the predominant monosaccharide across all EPS samples. Additionally, FTIR spectroscopy verified the presence of Amide I bands —indicative of protein components— alongside characteristic glycosidic bond vibrations. In terms of biological activity, the EPS derived from the C.vulgaris– B. subtilis co-culture demonstrated the highest antioxidant capacity, exhibiting a radical scavenging activity of 22.03%. Cytotoxicity analyses have shown that the combination of algal and bacterial EPS suppresses cancer cell proliferation more effectively and significantly than their pure forms. In conclusion, this study demonstrates that algal-bacterial interactions not only improve biomass yield but also significantly modulate the biological quality of the produced biopolymers, thereby enhancing their bioactive and therapeutic potential.
