ABSTRACT
OBJECTIVES: The
objektive of this study was to evaluated the differantitaion of human
adipose tissue derived mesenchymal stem cells (MSCs) into skeletal muscle cells
by the effect of IGF1 and FGF2 growth factors.
METHODS: MSCs
were divided into four groups. I.group (control group) was incubated only in
myogenic medium, II. group was incubated
by adding 100 ng/ml FGF2 to the myogenic medium, III. group was incubated by
adding 20 ng/ml IGF1 to the myogenic medium, IV. group was incubated by adding
100 ng/ml FGF2 and 20 ng/ml IGF1 to the myogenic medium. Cells dyed with MyoD1
antibody were analyzed by flow cytometry so as to determine myogenic
differentiation at the end of 28 days.
RESULTS: It
was confirmed that 11.5% of the control group, 39.2% of the FGF2 group, 23.1%
of the IGF1 group and 39.3% of the IGF1-FGF2 group showed differentiation.
CONCLUSION: Our
results show that IGF1 and FGF2 have a positive contribution on myogenic
differentiation of MSCs, contribute to the studies related to muscle diseases
and treatment of these diseases to support by growth factors the feature of the
regeneration capacity and differentiation.
