Kayseri yöresindeki hayvan barınaklarından toplanmış pirelerin morfolojik ve moleküler karakterizasyonları

Abstract:

This study was carried out to provide DNA barcoding by molecular characterization and establish phylogenetic constructions following the morphological identification of fleas collected from basement and shelter of building and animal barns exposed to flea infestation in Kayseri region. For this aim, a total of 100 flea samples were collected from animal barns and basement and shelter of building exposed to flea infestation between 2015-2016 in Kayseri region. The collected fleas were morphologically identified as Ctenocephalides canis in 45, as Ctenocephalides felis in 20 and as Pulex irritans in 35. Genomic DNA isolation from specimens was performed and PCR analyzes of the obtained isolates were done with the primer pairs that amplify the mt-COI gene region. The amplicons of 11 isolates belong to 5 for C. canis, 2 for C. felis and 4 for P. irritans, were cloned for sequence and phylogenetic analysis and plasmid purification was performed. The obtained plasmids were sequenced with the vector specific primer pairs and the sequences of the target gene regions were obtained. The molecular characterization of obtained isolate were determined, then they were deposited to GenBank (KY865409-19) and the phylogenetic analyses of the isolates recorded in GenBank (KY865409-19) were carried out by multiple alignments of the isolates with the similar flea ones available in GenBank in the world. According to mt-COI sequence analyses, genetic difference were determined as 0.1%, between C. canis isolates, 1.3% C. felis isolates and 2.6%. P. irritans isolates. In conclusion, the molecular characterizations and phylogenetic analyses of the fleas using together morphological and molecular analyses were firstly performed in Turkey with this study.
